Deafness is the most common sensory disorder in human affecting 1 in 1000 newborns. Most of these cases can be referred to genetic causes. Analyses in mice showed that mutations in deafness-related genes often result in morphological and functional abnormalities not only in the inner ear but also in the central auditory system (Satheesh et al., 2012; Willaredt et al., 2014).
Recent studies of our group identified components of the gene regulatory network to be essential for proper development and function of the auditory hindbrain (Schlueter et al., 2018; Ebbers et al., 2016). However, a comprehensive analysis of the genetic program underlying its formation is currently missing. To this end, we seek to apply state-of-the-art techniques such as single cell transcriptome analysis of fluorescent labeled cells to determine the gene regulatory network underlying the development of excitatory auditory neurons in the hindbrain. These neurons play a crucial role in sound localization.
- Isolation of fluorescent-labeled cells from brain tissue
- RNA-isolation for RNAseq analysis and RT-qPCR

- Mouse breeding and genotyping
We seek a highly motivated PhD student with:
- Academic university degree (Master or Diploma) in biology or related fields
- Hands-on experiences in cellular neurobiology or genetics (RNA isolation, PCR, immunohistochemistry)
- Experience in statistical analysis and microscopy
- Basic knowledge of gene regulatory networks or bioinformatics (Linux environment, R) are of advantage
- Experience with single cell isolation (FACS, patch-clamp, Laser microdissection) are of advantage
- Ability to work independently, thoroughly and as a team player in a young and dynamic group
